Because of the relatively low water solubility of the AM ester, the mild detergent Pluronic F-127 is often used as a dispersing agent to facilitate dye loading. Fura 2-AM (Fura 2) is a calcium-specific ratio-metric fluorescent dye. It is also known as acetoxy-methyl-ester. In response to calcium ions, ratio metric dyes like Fura 2 changes their respective excitation or emission spectra. The intracellular calcium concentration is determined by the fluorescence emission ratio or excitation at distinct wavelengths. Fura 2 has an emission peak at 505 nm, and the excitation peak ranges between 340 to 380 nm and is the preferred calcium indicator.

Xylene cyanol is often used as a tracking dye during agarose or polyacrylamide gel electrophoresis. It has a slightly negative charge and will migrate in the same direction as DNA, allowing the user to monitor the progress of molecules moving through the gel. The tracking dye typically migrates with the DNA molecules around 5 kb. Xylene cyanol may also be used in loading buffer that facilitates the observation of the sample during gel loading and electrophoresis.
